    Chloroquine CQ, which is frequently used clinically as an antimalarial agent, is a classic inhibitor of autophagy that blocks the binding of autophagosomes to lysosomes by altering the acidic environment of lysosomes, resulting in the accumulation of a large number of degraded proteins in cells. We show that autophagy is required for the most efficient degradation of glycogen in response to starvation. Chloroquine is a 4-aminoquinoline with antimalarial, anti-inflammatory, and potential chemosensitization and radiosensitization activities. Although the mechanism is not well understood, chloroquine is shown to inhibit the parasitic enzyme heme polymerase that converts the toxic heme into non-toxic hemazoin, thereby resulting in the accumulation of toxic heme within the parasite.

    Moreover, Chloroquine inhibits autophagy as it raises the lysosomal p H, which leads to inhibition of both fusion of autophagosome with lysosome and lysosomal protein degradation [4]. Chloroquine is commonly used to study the role of endosomal acidification in cellular processes [2, 3], such as the signaling of intracellular TLRs.
